You cannot access the files on your PC from the Netduino directly. You will need to provide your own functionality to do this. How to access a local file ex: "c:\test.txt" ?

If you are looking to provide an easy method of updating files on the SD card from your PC then there is a web server around on the forums which allows you use a web page to save a file to the SD card. This may help you achieve a better debug cycle for your pages.
Hope this help,
